I can see it being good as an option alongside the traditional square grid, but I think it would be too much of a departure from the traditional city designs people are used to creating if they made it the only option. Having hexagons as units can make it so that the awkward spaces get filled with buildings in a more natural way.

You forgot that not all cities are gridlike (acctualy it's in a 50/50), and even gridlike cities can get uneven parcels or plots. That's the thing, it's a game, not real life, and a system based on hexagons would be actually better to simulate the parcels. I can certainly see it being used for city expansion areas though. All lots being shaped like hexagons would look kind of odd.
